Pharmacokinetics

Pharmacokinetics of the parenteral form of Essentiale® N was studied by radioactive method. After intravenous administration single dose tritiated essential phospholipids (EPL), mainly containing linoleic acid, the incorporation of radioactivity into the fraction of cephalin, sphingomyelin and lysolecithin was noted. Among the neutral lipid fractions, labeled EPL appears to be more rapidly incorporated into free fatty acids than into cholesterol esters or triglycerides and are delivered to organs, mainly the liver. The uptake of the introduced lecithins by adipose tissue and erythrocytes was not detected. After intravenous administration, the half-life was 60 days. Less than 10% of the labeled substances were excreted in feces, through the kidneys and respiratory organs within 5-7 days after the dose was administered. It is assumed that the remainder of the tritiated essential phospholipids is transformed in the liver for the biosynthesis of HDL (high density cholesterol) or other lipoprotein fractions.

Pharmacodynamics

In liver diseases, the death of hepatocytes occurs, which leads to the release of liver enzymes and an increase in their activity in the blood serum.

The chemical structure of the phospholipids contained in Essentiale® N is similar to that of endogenous phospholipids, however, due to the high content of polyunsaturated (essential) fatty acids in them, the phospholipids in Essentiale® N are more active. The incorporation of high-energy phospholipid molecules into the structure of hepatic cell membranes increases the regeneration of hepatocyte membranes.

Due to the peculiarities of the chemical structure of essential phospholipids, the presence of cis-double bonds (arrangement of hydrocarbon chains on one side of double bonds) bonds in polyunsaturated fatty acids of cell membranes prevents the compaction of the structure of fatty acids. This leads to greater liquefaction of the membrane structure, which is necessary for the optimal functioning of most cells in the body. Through this mechanism, essential phospholipids act on those parts of cells that are receptors for hormones or neurotransmitters, thus increasing the metabolic rate, especially lipids. Phospholipids interfere with impaired lipid metabolism by regulating lipoprotein metabolism so that neutral fats and cholesterol are converted to transportable forms, and most importantly, this occurs by increasing the capacity of uptake of cholesterol by high density lipoprotein and can thus lead to its oxidation. Exogenous phospholipid molecules with their antioxidant properties and the ability to capture free radicals reliably protect lipids from peroxidation, that is, they suppress oxidative stress and reduce cell damage. Essential phospholipids reduce the production of hydrogen peroxide in hepatic microsomes and increase the concentration of reduced glutathione in the liver and plasma.

Intravenous administration of Essentiale® N solution improves the intestinal-hepatic circulation of bile acids in patients with impaired liver function. Phospholipids have a profound effect on the composition and flow of bile. During the excretion of phospholipids by bile, the risk of formation gallstones and the bile composition is stabilized.

Method of administration and dosage

Adults and children from 12 years old injected intravenously slowly, 5-10 ml, in severe cases, 10-20 ml per day. The maximum single dose is 10 ml, the maximum daily dose is 20 ml. The frequency of administration is 1-2 times a day.

Children from 3 to 6 years old2 ml per day, children from 6 to 12 years old, 2-5 ml per day.

Cannot be diluted in electrolyte solutions (saline sodium chloride solution, Ringer's solution, etc.).

"Essential" phospholipids optimally correspond to the chemical structure of human phospholipids. "Essential" phospholipids are incorporated into defective zones (gaps) of cell membranes, restoring their structure and metabolic activity. "Essential" phospholipids:

■ normalize liver function and enzymatic activity of liver cells;

■ reduce the level of energy consumption of the liver;

■ contribute to the regeneration (restoration) of the liver;

Indications for use

ESSENTIALE N is a preparation made from raw materials vegetable originused to treat liver disease.

It is used in patients with the various liver disorders listed below to relieve symptoms such as loss of appetite and / or a feeling of heaviness in the right hypochondrium, which may indicate the following liver damage and / or disease:

Fatty liver degeneration (also in diabetes);

Contraindications

Method of administration and dosage

Contraindications

Besides traditional methods application, Essentiale solution is used and not quite standard. One option is to use it to strengthen hair.

  • To strengthen the hair follicles, it is necessary to make a mask from the preparation added to a mixture of egg yolks with sour cream (or kefir).
  • For a mask for short hair, it is enough to use a third of the solution from one ampoule. On long hair - half an ampoule.
  • The preparation of the healing strengthening composition is very simple: having collected the solution with a syringe, it is mixed with the rest of the ingredients (1 yolk and 1-2 tsp / l sour cream or kefir).
  • Apply the mixture to washed and slightly dried hair. Keep the mask on your hair for an hour. After that, it is washed off with water without shampoo.

The frequency of the procedure is once a week. The course is 5 weeks. Subsequently, the mask can be applied for prophylaxis once a month.

Important. Dimexide will help to enhance the effect of the strengthening mixture.

Rules for using Essentiale solution

For the drug to bring maximum benefit, it must be used correctly. To do this, you should adhere to some rules:

  1. Inject only with clear solution. If, during mixing or during the introduction, the slightest signs of turbidity appear in it, then the procedure must be interrupted, discard the container with the cloudy contents. The rest of the dose of the drug must be administered from another ampoule.
  2. Essentiale and other drugs cannot be combined in one syringe.
  3. No normal saline solution is used to dilute the drug. It should be diluted with the patient's own blood in a 1: 1 ratio. Instead of blood, it is allowed to use electrolyte-free solutions (5% xylitol, 5 or 10% glucose). An exception is made only when blood cannot be used.
  4. The drug is administered exclusively intravenously. It cannot be used intramuscularly. Also, you can not make subcutaneous injections, since there is a high probability of irritation at the injection site.
  5. The medicine is administered very slowly.
  6. If the patient is prescribed a large number of injections, then the injections are replaced with droppers.
